Applications Validated:
WB: 1-2ug/ml dilution
For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all blocking solutions may also reduce background.
Immunogen
This MBD4 antibody was raised against a mixture of synthetic peptides corresponding to amino acid residues 268-282 and 337-352 of human MBD4.
Preparation and Storage
Some products may be shipped at room temperature. This will not affect their stability or performance. Avoid repeated freeze/thaw cycles by aliquoting items into single-use fractions for storage at -20 degree C for up to 2 years. Keep all reagents on ice when not in storage.1ug/ul

Related Product Information for anti-MBD4 antibody
Short Description: Our MBD4 antibody (pAb) was raised in a Rabbit host. It has been validated for use in Western blot; it has been shown to react with Human samples.

Background: Methylation of mammalian DNA has long been recognized to play a major role in a number of cellular functions such as development and control of gene expression. It is generally associated with the repressive chromatin state. The complex series of events leading to this repressive state involve the coordinated regulation of DNA methyltransferases and two other groups of proteins called the Methyl-CpG binding proteins (MBD proteins) and the Kaiso family of proteins. The MBD family of proteins include MeCP2, MBD1, MBD2, MBD3 and MBD4. MBD4 (methyl-CpG binding domain protein 4) is a thymine glycosylase that recognizes the product of deamination at methyl-CpG sites, as a part of DNA repair system. MBD4 is able to bind the hemimethylated DNA or methyl-CpG TpG mismatches.

NCBI Description

The protein encoded by this gene is a member of a family of nuclear proteins related by the presence of a methyl-CpG binding domain (MBD). These proteins are capable of binding specifically to methylated DNA, and some members can also repress transcription from methylated gene promoters. This protein contains an MBD domain at the N-terminus that functions both in binding to methylated DNA and in protein interactions and a C-terminal mismatch-specific glycosylase domain that is involved in DNA repair. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Jan 2013]

Uniprot Description

MBD4: Mismatch-specific DNA N-glycosylase involved in DNA repair. Has thymine glycosylase activity and is specific for G:T mismatches within methylated and unmethylated CpG sites. Can also remove uracil or 5-fluorouracil in G:U mismatches. Has no lyase activity. Was first identified as methyl-CpG-binding protein. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The MBD4 mbd4 (Catalog #AAA388413) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The MBD4 antibody (pAb) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's MBD4 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Applications Validated: WB: 1-2ug/ml dilution For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all blocking solutions may also reduce background. Researchers should empirically determine the suitability of the MBD4 mbd4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MBD4, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
