Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Type I insulin-like growth factor receptor (IGF-IR) is a transmembrane tyrosine kinase. It is widely expressed among many cell types in fetal and postnatal tissues and many cell lines. Upon binding of its ligands, IGF-I and IGF-II, autophosphorylation of receptors occurs. The triple tyrosine cluster (Tyr1131, Tyr1135 and Tyr1136) within the kinase domain is the earliest major site of autophosphorylation. Phosphorylation of these three tyrosine residues is necessary for activation of kinases. Insulin receptors (IRs) share significant similarity with IGF-I receptors in structure and function. There is also an equivalent triple tyrosine cluster (Tyr1146, Tyr1150 and Tyr1151) in its activation loop of the kinase domain. Tyrosine autophosphorylation of the receptors is one of the earliest cellular responses to insulin stimulation. The autophosphorylation begins by phosphorylation of Tyr1146 and either Tyr1150 or Tyr1151. The full activation of receptor kinase activity depends on the triple tyrosine phosphorylation.

IRS4 encodes the insulin receptor substrate 4, a cytoplasmic protein that contains many potential tyrosine and serine/threonine phosphorylation sites. Tyrosine-phosphorylated IRS4 protein has been shown to associate with cytoplasmic signalling molecules that contain SH2 domains. The IRS4 protein is phosphorylated by the insulin receptor tyrosine kinase upon receptor stimulation.. [provided by RefSeq, Jul 2008]

IRS4: Acts as an interface between multiple growth factor receptors possessing tyrosine kinase activity, such as insulin receptor, IGF1R and FGFR1, and a complex network of intracellular signaling molecules containing SH2 domains. Involved in the IGF1R mitogenic signaling pathway. Promotes the AKT1 signaling pathway and BAD phosphorylation during insulin stimulation without activation of RPS6KB1 or the inhibition of apoptosis. Interaction with GRB2 enhances insulin-stimulated mitogen-activated protein kinase activity. May be involved in nonreceptor tyrosine kinase signaling in myoblasts. Plays a pivotal role in the proliferation/differentiation of hepatoblastoma cell through EPHB2 activation upon IGF1 stimulation. May play a role in the signal transduction in response to insulin and to a lesser extent in response to IL4 and GH on mitogenesis. Plays a role in growth, reproduction and glucose homeostasis. May act as negative regulators of the IGFI signaling pathway by suppressing the function of IRS1 and IRS2.
