Application Notes
ELISA: This purified mouse IL-12 antibody may be used in an indirect ELISA or as the capture reagent in a sandwich ELISA with a biotinylated mouse IL-12 antibody as the detection reagent and recombinant mouse IL-12 as the standard.
Western Blot: This purified mouse IL-12 antibody may used in Western Blotting applications under either reducing or non-reducing conditions with recombinant mouse IL-12 as the positive control.
ELISA: Minimum Dilution: 0.5; Maximum Dilution: 2.0 ug/ml
Functional Assays: Minimum Dilution: 2.0; Maximum Dilution: 3.0 ng/ml
Western Blotting: Minimum Dilution: 0.1; Maximum Dilution: 0.2 ug/ml
Reconstitution
Reconstitute with 0.1ml distilled water Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution. For long term storage the addition of 0.09% sodium azide is recommended. N.B. For functional studies do not add sodium azide

Antisera to mouse IL-12 were raised by repeated immunisations of goats with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.

Preparation and Storage
Prior to reconstitution store at 4 degree C. Following reconstitution store at -20 degree C. This product should be stored undiluted. Storage in frost-free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of reconstitution.

Goat anti Mouse Interleukin-12 antibody recognizes mouse interleukin-12 (IL-12). Interleukin-12 is produced by activated B lymphocytes and macrophages. The properties of IL-12 include the stimulation of growth of activated T lymphocytes and the development of TH1 CD4 +ve cells and CD8 +ve cytotoxic T cells. Goat anti Mouse Interleukin-12 antibody neutralizes murine IL-12 activity with an ND50 for mIL-12 (2ngml-1 of between 0.002 and 0.002μg ml-1.

This gene encodes the beta subunit p40 of the Interleukin 12 (IL-12) family of cytokines. Members of the IL-12 family form heterodimers consisting of heavy and light subunits linked by disulfide bonds. The product of this gene, p40, is a subunit of interleukins IL-12 and IL-23. IL12B: Cytokine that can act as a growth factor for activated T and NK cells, enhance the lytic activity of NK/lymphokine- activated killer cells, and stimulate the production of IFN-gamma by resting PBMC. Defects in IL12B are a cause of mendelian susceptibility to mycobacterial disease (MSMD); also known as familial disseminated atypical mycobacterial infection. This rare condition confers predisposition to illness caused by moderately virulent mycobacterial species, such as Bacillus Calmette-Guerin (BCG) vaccine and environmental non-tuberculous mycobacteria, and by the more virulent Mycobacterium tuberculosis. Other microorganisms rarely cause severe clinical disease in individuals with susceptibility to mycobacterial infections, with the exception of Salmonella which infects less than 50% of these individuals. The pathogenic mechanism underlying MSMD is the impairment of interferon-gamma mediated immunity, whose severity determines the clinical outcome. Some patients die of overwhelming mycobacterial disease with lepromatous-like lesions in early childhood, whereas others develop, later in life, disseminated but curable infections with tuberculoid granulomas. MSMD is a genetically heterogeneous disease with autosomal recessive, autosomal dominant or X-linked inheritance. Genetic variations in IL12B are a cause of susceptibility to psoriasis type 11 (PSORS11). Psoriasis is a common, chronic inflammatory disease of the skin with multifactorial etiology. It is characterized by red, scaly plaques usually found on the scalp, elbows and knees. These lesions are caused by abnormal keratinocyte proliferation and infiltration of inflammatory cells into the dermis and epidermis. Belongs to the type I cytokine receptor family. Type 3 subfamily.
