Rabbit anti-Human HPC2 / CBX4 Polyclonal Antibody | anti-HPC2 antibody
Rabbit Polyclonal to Human HPC2 / CBX4
Usage: Immunohistochemistry: was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antib...
Immunohistochemistry (IHC)
(Anti-CBX4 antibody IHC of human colon. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody concentration 5 ug/ml.)
Immunofluorescence (IF)
(Anti-hPC2 Antibody - Immunofluorescence Microscopy. anti-hPC2 antibody was used for immunofluorescent imaging of human cells (U2OS). The image reveals the expected discrete nuclear structure that is termed the PcG body corresponding to the known localization of PC2 (see Satijn et al. below). IF was performed after fixation in PBS with 4% PF for 5 min, permeabilization with 0.5% Triton X100-PBS for 5 min, and blocking with 5% milk / 0.2% Tween for 1 h. Primary antibody used at 1:200 in 5% milk / 0.2% Tween for 1 h, secondary antibody for 30 min. All blocking and incubation steps carried out at 37° C. Nuclei were counterstained with Hoechst stain (blue). Data contributed by Luke Hughes-Davies and Rhiannon Jade, Gurdon Institute, Cambridge, UK.)
Western Blot (WB)
(Anti-hPC2 Antibody - Western Blot. Analysis shows the detection of human PC2 in probed lysates using Affinity Purified anti-hPC2 antibody.)
NCBI and Uniprot Product Information
Uniprot Description
CBX4: E3 SUMO-protein ligase which facilitates SUMO1 conjugation by UBE2I. Involved in the sumoylation of HNRNPK, a p53/TP53 transcriptional coactivator, hence indirectly regulates p53/TP53 transcriptional activation resulting in p21/CDKN1A expression. 2 isoforms of the human protein are produced by alternative splicing.
Protein type: Transcription, coactivator/corepressor; EC 6.3.2.-; SUMO ligase
Chromosomal Location of Human Ortholog: 17q25.3
Cellular Component: nucleoplasm; nuclear speck; PcG protein complex; nucleus
Molecular Function: protein binding; enzyme binding; single-stranded RNA binding; SUMO binding; phosphoprotein binding; chromatin binding; transcription corepressor activity; methylated histone residue binding; ligase activity

The HPC2 cbx4 (Catalog #AAA240839) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Rabbit Polyclonal to Human HPC2 / CBX4 reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's HPC2 / CBX4 can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC - Paraffin), Immunofluorescence (IF), Western Blot (WB), ELISA (EIA). ELISA (1:10000 - 1:40000), IF (1:100 - 1:400), IHC-P (2.5 ug/ml), WB (1:1000 - 1:4000) Usage: Immunohistochemistry: was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antib.. Researchers should empirically determine the suitability of the HPC2 cbx4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "HPC2 / CBX4, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
