Applications are based on unconjugated antibody.
Western Blot (WB)
(Western blot analysis of GZMB Antibody (C-term) in HL-60 cell line lysates (35ug/lane). GZMB (arrow) was detected using the purified Pab.)
Immunohistochemistry (IHC)
(GZMB Antibody (C-term) IHC analysis in formalin fixed and paraffin embedded human Lymph tissue followed by peroxidase conjugation of the secondary antibody and DAB staining. This data demonstrates the use of the GZMB Antibody (C-term) for immunohistochemistry. Clinical relevance has not been evaluated.)
Flow Cytometry (FC/FACS)
(GZMB Antibody (C-term) flow cytometric analysis of HL-60 cells (right histogram) compared to a negative control cell (left histogram).FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.)
Western Blot (WB)
(Western blot analysis of GZMB (arrow) using rabbit polyclonal GZMB Antibody (C-term). 293 cell lysates (2 ug/lane) either nontransfected (Lane 1) or transiently transfected (Lane 2) with the GZMB gene.)
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a member of the granzyme subfamily of proteins, part of the peptidase S1 family of serine proteases. The encoded preproprotein is secreted by natural killer (NK) cells and cytotoxic T lymphocytes (CTLs) and proteolytically processed to generate the active protease, which induces target cell apoptosis. This protein also processes cytokines and degrades extracellular matrix proteins, and these roles are implicated in chronic inflammation and wound healing. Expression of this gene may be elevated in human patients with cardiac fibrosis. [provided by RefSeq, Sep 2016]
Uniprot Description
GZMB: This enzyme is necessary for target cell lysis in cell- mediated immune responses. It cleaves after Asp. Seems to be linked to an activation cascade of caspases (aspartate-specific cysteine proteases) responsible for apoptosis execution. Cleaves caspase-3, -7, -9 and 10 to give rise to active enzymes mediating apoptosis. By staphylococcal enterotoxin A (SEA) in peripheral blood leukocytes. Inactivated by the serine protease inhibitor diisopropylfluorophosphate. Belongs to the peptidase S1 family. Granzyme subfamily.

The Granzyme B, CT (GRB, GZMB, C11, Cathespin G-like 1, CCPI, CGL1, CGL-1, CSPB, CSP-B, CTLA1, CTLA-1, CTSGL1, Cytotoxic T Lymphocyte-associated Serine Esterase 1, Cytotoxic T Lymphocyte Proteinase 2, Fragmentin 2, Fragmentin-2, Granzyme 2, Human Lymphocyte P reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Granzyme B can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), FLISA. WB: 1:50-1:100 FLISA: 1:1,000 Applications are based on unconjugated antibody. Researchers should empirically determine the suitability of the GZMB gzmb for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Granzyme B, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
