This antibody is specific for rat glutamate receptor subunit 1 (GluR1) when phosphorylated at serine 831. GluR1, widely expressed throughout the nervous system, is a component of AMPA receptors (alpha-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid receptors), which play a key role at excitatory synapses. Phosphorylation of Ser831 and Ser845 is reported to potentiate GluR1 function, suggesting a possible role in synaptic plasticity, learning and memory.

Western Blot: This item detects a band of approximately 100 kDa in rat brain lysates.
Western Blotting: Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.

Antisera to phosphorylated GluR1 were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared by sequential affinity chromatography on phospho- and dephosphopeptide affinity columns.

Store at 4 degree C or -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

GluR1: an integral membrane protein belonging to the glutamate-gated ion channel family. L-glutamate (Glu) acts as an excitatory neurotransmitter at many synapses in the central nervous system. Glutamate receptors are heteromeric protein complexes with multiple subunits, each possessing transmembrane regions, and all arranged to form a ligand-gated ion channel. The postsynaptic actions of Glu are mediated by a variety of receptors that are named according to their selective agonists. Each of the four GluR proteins (GRIA1-4) include flip and flop isoforms generated by alternative RNA splicing. Expressed in granule and pyramidal cells in the hippocampal formation.
