Western Blot (WB)
(Western blot analysis of extracts from Mouse brain , using Epac2 Antibody. The lane on the left was treated with blocking peptide.)
Immunofluorescence (IF)/Immunocytochemistry (ICC)
(MBS9612301 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612301 1:200) and mouse anti-beta tubulin Ab(T0023 1:200) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
Subcellular Location: Cytoplasm. Membrane>Peripheral membrane protein.
Tissue Specificity: Predominantly expressed in brain and adrenal gland. Isoform 2 is expressed in liver. Isoform 1 is expressed in liver at very low levels.
Subunit Structure: Interacts with RIMS1 and RIMS2. Probably part of a complex with RIMS2 and GTP-activated RAB3A (By similarity).
Similarity: The N-terminal nucleotide phosphate binding region cAMP 1 has a much lower affinity for cAMP as compared to cAMP 2.The DEP domain is involved in membrane localization independent from regulation by cAMP.
NCBI and Uniprot Product Information
Predicted Molecular Weight: (Calculated)116kDa.
Uniprot Description
RAPGEF4: Guanine nucleotide exchange factor (GEF) for RAP1A, RAP1B and RAP2A small GTPases that is activated by binding cAMP. Seems not to activate RAB3A. Involved in cAMP-dependent, PKA- independent exocytosis through interaction with RIMS2. Interacts with RIMS1 and RIMS2. Probably part of a complex with RIMS2 and GTP-activated RAB3A. Predominantly expressed in brain and adrenal gland. Isoform 2 is expressed in liver. Isoform 1 is expressed in liver at very low levels. 3 isoforms of the human protein are produced by alternative promoter.

Product Notes
The RAPGEF4 rapgef4 (Catalog #AAA9612301) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Epac2 Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Pig(100%), Bovine(100%), Horse(100%), Sheep(100%), Rabbit(88%), Dog(100%) and may cross-react with other species as described in the data sheet. AAA Biotech's Epac2 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). IF: 1:100-1:500 ICC: 1:100-1:500 WB: 1:500-1:2000 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the RAPGEF4 rapgef4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Epac2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
