Biological Significance
DBH catalyzes the conversion of dopamine to norepinephrine and serves as a marker of noradrenergic cells. DBH antibodies and antibodies for other markers of catecholamine biosynthesis are widely used as markers for dopaminergic and noradrenergic neurons in a variety of applications including depression, schizophrenia, Parkinson's disease and drug abuse (Kish et al., 2001; Zhu et al., 2000; Zhu et al., 1999). The expression of DBH is also elevated during stress (Sabban and Kvetnansky, 2001).
Preparation and Storage
For long term storage -20 degree C is recommended. Stable at -20 degree C for at least 1 year.

NCBI Description

The protein encoded by this gene is an oxidoreductase belonging to the copper type II, ascorbate-dependent monooxygenase family. It is present in the synaptic vesicles of postganglionic sympathetic neurons and converts dopamine to norepinephrine. It exists in both soluble and membrane-bound forms, depending on the absence or presence, respectively, of a signal peptide. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Conversion of dopamine to noradrenaline.

Catalytic activity: 3,4-dihydroxyphenethylamine + ascorbate + O2 = noradrenaline + dehydroascorbate + H2O.

Cofactor: Binds 1 PQQ per subunit.Binds 2 copper ions per subunit.

Pathway: Catecholamine biosynthesis; (R)-noradrenaline biosynthesis; (R)-noradrenaline from dopamine: step 1/1.

Subunit structure: Homotetramer composed of two non-covalently bound disulfide-linked dimers.

Subcellular location: Soluble dopamine beta-hydroxylase: Cytoplasmic vesicle › secretory vesicle lumen. Cytoplasmic vesicle › secretory vesicle › chromaffin granule lumen. Cytoplasmic vesicle › secretory vesicle membrane; Single-pass type II membrane protein. Cytoplasmic vesicle › secretory vesicle › chromaffin granule membrane; Single-pass type II membrane protein

Potential.

Induction: Activity is enhanced by nerve growth factor (in superior cervical ganglia and adrenal medulla). Trans-synaptic stimulation with reserpine, acetylcholine and glucocorticoids.

Polymorphism: There are two main alleles of DBH: DBH-A with Ala-318 and DBH-B with Ser-318.

Involvement in disease: Dopamine beta-hydroxylase deficiency (DBH deficiency) [MIM:223360]: Characterized by profound deficits in autonomic and cardiovascular function, but apparently only subtle signs, if any, of central nervous system dysfunction.Note: The disease is caused by mutations affecting the gene represented in this entry.

Sequence similarities: Belongs to the copper type II ascorbate-dependent monooxygenase family.Contains 1 DOMON domain.

Sequence caution: The sequence AAH17174.1 differs from that shown. Reason: Erroneous initiation. The sequence CAA31631.1 differs from that shown. Reason: Erroneous initiation. The sequence CAA31632.1 differs from that shown. Reason: Erroneous initiation. The sequence CAA68285.1 differs from that shown. Reason: Erroneous initiation.

Product Notes

The DBH dbh (Catalog #AAA502023) is an Antibody produced from Sheep and is intended for research purposes only. The product is available for immediate purchase. The Anti-Dopamine beta-Hydroxylase, C-Terminus reacts with Human, mouse, and non-human primate and may cross-react with other species as described in the data sheet. AAA Biotech's Dopamine beta-Hydroxylase, C-Terminus, Human can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Quality Control: Western blots performed on each lot. WB: 1:1000. Researchers should empirically determine the suitability of the DBH dbh for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Dopamine beta-Hydroxylase, C-Terminus, Human, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
