May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Deoxyribonuclease II (Dnase II, EC.3.1.22.1) is one of the two Dnase present in mammalian tissues and biological fluids. Dnase II hydrolyzes DNA to 3'-phosphoryl oligonucleotides under acidic conditions. Dnase II is also known as "acid Dnase". It has preference of for dsDNA. Dnase II has been implicated in internucleosomal cleavage of chromatin to generate DNA fragments or ladder, a hallmark of apoptosis. Dnase II (mouse 353aa; human 360aa, chromosome 19p13.2-q13.4) is a lysosomal enzyme present in most tissues. Dnase Ii is synthesized as a pro-enzyme (signal peptide aa1-16, propeptide region of 91-aa, and 253-aa mature protein (aa110-353) and processed to produce active enzyme containing 2- chains of ~32 and 12kD. Dnase II has no significant sequence homology with Dnase I.

NCBI Description

This gene encodes a member of the DNase family. The protein, located in the lysosome, hydrolyzes DNA under acidic conditions and mediates the breakdown of DNA during erythropoiesis and apoptosis. Two codominant alleles have been characterized, DNASE2*L (low activity) and DNASE2*H (high activity), that differ at one nucleotide in the promoter region. The DNASE2*H allele is represented in this record. [provided by RefSeq]

DNASE2: Hydrolyzes DNA under acidic conditions with a preference for double-stranded DNA. Plays a major role in the degradation of nuclear DNA in cellular apoptosis during development. Necessary for proper fetal development and for definitive erythropoiesis in fetal liver, where it degrades nuclear DNA expelled from erythroid precursor cells. Belongs to the DNase II family.
