Related Product Information for anti-DAP12 antibody
Background: DAP12 is a transmembrane adaptor protein that is non-covalently associated with several cell surface receptors on natural killer (NK), myeloid, and presumably neuronal cells (1,2). DAP12 contains an ITAM domain that preferentially recruits the protein tyrosine kinase Syk to initiate signal transduction (3). Deficiency of DAP12 results in reduced antigen presentation function by myeloid cells and defects in function of certain NK cell receptors in mice (4,5), as well as presenile dementia and bone cysts in humans (6).

Uniprot Description

Function: Non-covalently associates with activating receptors of the CD300 family. Cross-linking of CD300-TYROBP complexes results in cellular activation. Involved for instance in neutrophil activation mediated by integrin. Ref.7

Subunit structure: Homodimer; disulfide-linked. Interacts with SIRPB1 and TREM1. Interacts with SIGLEC14

By similarity. Interacts with CLECSF5. Interacts with CD300LB and CD300D. Interacts with CD300E. Interacts with KLRC2 and KIR2DS3

By similarity. Interacts (via ITAM domain) with SYK (via SH2 domains); activates SYK mediating neutrophils and macrophages integrin-mediated activation. Ref.4 Ref.5 Ref.6 Ref.7 Ref.8 Ref.10

Subcellular location: Membrane; Single-pass type I membrane protein.

Post-translational modification: Tyrosine phosphorylated.

Sequence similarities: Belongs to the TYROBP family.Contains 1 ITAM domain.

Product Notes

The DAP12 tyrobp (Catalog #AAA395757) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The DAP12 reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's DAP12 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunoprecipitation (IP). Antibody can be used for immunoprecipitation (2-4 ug/mL protein lysate)
Optimal concentration should be evaluated by serial dilutions. Researchers should empirically determine the suitability of the DAP12 tyrobp for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "DAP12, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
