Intended Uses: This sandwich kit is for the accurate quantitative detection of human Chitinase1 (also known as CHIT1) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.
Principle of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human CHIT1 antibody. CHIT1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human CHIT1 Antibody is added and binds to CHIT1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CHIT1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human CHIT1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Chitotriosidase is secreted by activated human macrophages and is markedly elevated in plasma of Gaucher disease patients. The expression of chitotriosidase occurs only at a late stage of differentiation of monocytes to activated macrophages in culture. Human macrophages can synthesize a functional chitotriosidase, a highly conserved enzyme with a strongly regulated expression. This enzyme may play a role in the degradation of chitin-containing pathogens. Several alternatively spliced transcript variants have been described for this gene. [provided by RefSeq, Jan 2012]
