Post Translational Modifications
Phosphorylation/dephosphorylation states of Ser-273 are required for regulating transduction of CREB activity (By similarity). CRTCs/TORCs are inactive when phosphorylated, and active when dephosphorylated at this site (By similarity). May be phosphorylated at Ser-391 by MAPK3/ERK1 and/or MAPK1/ERK2 or by some cyclin-dependent kinases such as CDK1,CDK2 or CDK5. Following adenylyl cyclase activation, dephosphorylated at Ser-162 and Ser-273 resulting in its dissociation from 14-3-3 proteins probably promoting CRTC3 translocation into the nucleus.
Subunit Structure
Binding, as a tetramer, through its N-terminal region, with the bZIP domain of CREB1 enhances recruitment of TAF4 to the promoter. 'Arg-314' in the bZIP domain of CREB1 is essential for this interaction. Interacts (when phosphorylated at Ser-162 and Se-273) with 14-3-3 proteins. Interacts with YWHAE. Interacts (when phosphorylated at Ser-391) with phosphatase PP2A catalytic subunit PPP2CA and regulatory subunits PPP2R1A and PPP2R2A. Interacts, via the N-terminal with the ankyrin repeats of BCL3, to form a complex with CREB1 on CRE and TxRE responsive elements and represses HTLV-1 LTR-mediated transcription. (Microbial infection) Interacts with HTLV-1 protein Tax; this interaction enhances tax transcriptional activity.
Similarity
Belongs to the TORC family.
Subcellular Location
Nucleus. Cytoplasm.
Note: Appears to be mainly nuclear (PubMed:15454081). Translocates to the nucleus following adenylyl cyclase or MAP kinase activation (PubMed:30611118).
Preparation and Storage
Store at -20 degree C. Stable for 12 months from date of receipt.

Related Product Information for anti-CRTC3 antibody
Transcriptional coactivator for CREB1 which activates transcription through both consensus and variant cAMP response element (CRE) sites. Acts as a coactivator, in the SIK/TORC signaling pathway, being active when dephosphorylated and acts independently of CREB1 'Ser-133' phosphorylation. Enhances the interaction of CREB1 with TAF4. Regulates the expression of specific CREB-activated genes such as the steroidogenic gene, StAR. Potent coactivator of PPARGC1A and inducer of mitochondrial biogenesis in muscle cells. Also coactivator for TAX activation of the human T-cell leukemia virus type 1 (HTLV-1) long terminal repeats (LTR).

NCBI Description

This gene is a member of the CREB regulated transcription coactivator gene family. This family regulates CREB-dependent gene transcription in a phosphorylation-independent manner and may be selective for cAMP-responsive genes. The protein encoded by this gene may induce mitochondrial biogenesis and attenuate catecholamine signaling in adipose tissue. A translocation event between this gene and Notch coactivator mastermind-like gene 2, which results in a fusion protein, has been reported in mucoepidermoid carcinomas. Alternative splicing results in multiple transcript variants that encode different protein isoforms. [provided by RefSeq, Jul 2012]

Product Notes

The CRTC3 crtc3 (Catalog #AAA9613598) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The CRTC3 Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's CRTC3 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Peptide ELISA (EIA). WB: 1:500-1:2000 Peptide ELISA: 1:20,000-1:40,000. Researchers should empirically determine the suitability of the CRTC3 crtc3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CRTC3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
