Specificity
This item specifically recognises 43kDa Connexin43, when phosphorylated at Ser368. Connexin43 (CX43) allows for the exchange of solutions between neighbouring cells through the formation of gap-junctional channels and is expressed in the brain, kidneys and myocardium, as well as vascular endothelial cells. Studies suggest that a decrease in the permeability of gap-junctional channels occurs due to a conformational change in the structure of CX43, and that this is mediated by the phosphorylation of CX43 at residue Ser368, by Protein Kinase C (PKC). A possible link between CX43 and epilepsy has been investigated following the detection of high levels of CX43 in both low-grade tumours and peritumoural reactive astrocytes, see Aronica, E. et al. for details.

Applicable Applications for anti-Gja1 antibody
Western Blot (WB)*
Application Notes
Western Blot: This item detects a band of approximately 43kDa in rat hippocampus cell lysates.
Western Blotting: Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
Perservative Stabilisers
0.09% Sodium Azide
0.01% Bovine Serum Albumin
50% Glycerol
Immunogen
Synthetic phosphopeptide corresponding to an amino acid sequence within connexin43 which includes phosphorylated Ser368
Buffer Solution
Antiserum Preparation
Antisera to rat connexin43 were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared by affinity chromatography.
Target Species
Rat
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of despatch.

NCBI Description

gap junction component; plays a role in cell-cell communication [RGD, Feb 2006]

Uniprot Description

GJA1: an integral membrane protein of the connexin family, alpha-type (group II) subfamily. Hexamers of connexin-43 form connexons, which aggregate together to form gap junctions, through which materials of low MW diffuse from one cell to a neighboring cell. May play a critical role in the physiology of hearing by participating in the recycling of potassium to the cochlear endolymph.

Product Notes

The Gja1 gja1 (Catalog #AAA223268) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI CONNEXIN 43 (pSer368) reacts with Bovine (Expected from Sequence), Chicken (Expected from Sequence), Dog (Expected from Sequence), Guinea Pig (Expected from Sequence), Human (Expected from Sequence), Mouse (Expected from Sequence), Zebrafish (Expected from Sequence) and may cross-react with other species as described in the data sheet. AAA Biotech's CONNEXIN 43 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB)*. Researchers should empirically determine the suitability of the Gja1 gja1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CONNEXIN 43, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
