By Heat: Boiling the paraffin sections in 10mM citrate buffer, pH6.0, for 20mins is required for the staining of formalin/paraffin sections.
Other applications have not been tested.
Reconstitution
Add 0.2ml of distilled water will yield a concentration of 500ug/ml.
Immunogen
E. coli-derived mouse CD40/TNFRSF5 recombinant protein (Position: L20-R193). Mouse CD40/TNFRSF5 shares 60.7% amino acid (aa) sequence identity with human CD40/TNFRSF5.
Preparation and Storage
Store at -20 degree C for one year. After reconstitution, at 4 degree C for one month. It can also be aliquotted and stored frozen at -20 degree C for a longer time. Avoid repeated freezing and thawing.

(CD40/TNFRSF5 was detected in paraffin-embedded sections of mouse lymphaden tissues using rabbit anti- CD40/TNFRSF5 Antigen Affinity purified polyclonal antibody at 1 ug/mL. The immunohistochemical section was developed using SABC method. )

Isotype control antibody (Green line) was rabbit IgG (1ug/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control. )

Background: CD40 is a cell surface receptor that is expressed on the surface of all mature B cells and also expressed on monocytes, dendritic cells, and thymic epithelium. It is a member of the tumor necrosis factor (TNF) receptor superfamily and is the receptor for CD40 ligand. CD40 ligand (CD40L, CD154, gp39, and TRAM) belongs to the TNF gene family and is expressed more widely than CD40 predominantly on activated CD4+ T cells. Activation of CD40 has also been shown to inhibit the growth of certain B cell lymphomas and to induce the death of transformed cells of mesenchymal or epithelial origin.

Uniprot Description

CD40: a member of the TNF-receptor superfamily. This receptor for CD40L mediates a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. Defects in CD40 are the cause of hyper-IgM immunodeficiency type 3 (HIGM3). HIGM3 is an autosomal recessive disorder which includes an inability of B cells to undergo isotype switching, one of the final differentiation steps in the humoral immune system, an inability to mount an antibody-specific immune response, and a lack of germinal center formation. Two alternatively spliced isoforms have been reported. Isoform I is a type I membrane protein; isoform II is secreted.
