Background: The acetylation of histone lysine residues plays a crucial role in the epigenetic regulation of gene transcription. A bromodomain is a protein domain that recognizes acetylated lysine residues such as those on the N-terminal tails of histones. This recognition is often a prerequisite for protein-histone association and chromatin remodeling. These domains function in the linking of protein complexes to acetylated nucleosomes, thereby controlling chromatin structure and gene expression. Thus, bromodomains serve as "readers" of histone acetylation marks regulating the transcription of target promoters. The BET family of proteins, defined by tandem Bromodomains and an Extra Terminal domain, include BRD2, BRD3, BRD4, and BRDT. The BET proteins play a key role in many cellular processes, including inflammatory gene expression, mitosis, and viral/host interactions. The isolated individual or tandem bromodomains of BRD3 have been shown to bind acetylated histone tails, serving to couple histone acetylation marks to the transcriptional regulation of target promoters. Small molecule inhibitors of these interactions hold promise as useful therapeutics for human disease.

NCBI Description

This gene was identified based on its homology to the gene encoding the RING3 protein, a serine/threonine kinase. The gene localizes to 9q34, a region which contains several major histocompatibility complex (MHC) genes. The function of the encoded protein is not known. [provided by RefSeq, Jul 2008]

Uniprot Description

BRD3: Binds hyperacetylated chromatin and plays a role in the regulation of transcription, probably by chromatin remodeling and interaction with transcription factors. Regulates transcription by promoting the binding of the transcription factor GATA1 to its targets. Regulates transcription of the CCND1 gene. A chromosomal aberration involving BRD3 is found in a rare, aggressive, and lethal carcinoma arising in midline organs of young people. Translocation t(15;9)(q14;q34) with NUT which produces a BRD3-NUT fusion protein. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Protein kinase, atypical; Protein kinase, Ser/Thr (non-receptor); Kinase, protein; ATYPICAL group; BRD family

Chromosomal Location of Human Ortholog: 9q34

Cellular Component: nucleus

Molecular Function: protein binding; chromatin binding

Biological Process: regulation of transcription from RNA polymerase II promoter; transcription, DNA-dependent; chromatin modification

Product Notes

The BRD3 brd3 (Catalog #AAA840422) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The BRD3 Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's BRD3 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). WB: 1:200 Note: This information is only intended as a guide. The optimal dilutions must be determined by the use. Researchers should empirically determine the suitability of the BRD3 brd3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "BRD3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
