Immunohistochemistry (IHC)
(Fig: 3 Formalin-fixed, paraffin-embedded normal human colon epithelium tissue section stained for Bcl-X expression using at 1: 2000. Hematoxylin-eosin counterstain. A course granular Bcl-X staining pattern associated with the mitochondria is observed.)
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a member of the BCL-2 family. BCL-2 family members are known to be regulators of programmed cell death. This protein positively regulates cell apoptosis by forming heterodimers with BCL-xL and BCL-2, and reversing their death repressor activity. Proapoptotic activity of this protein is regulated through its phosphorylation. Protein kinases AKT and MAP kinase, as well as protein phosphatase calcineurin were found to be involved in the regulation of this protein. Alternative splicing of this gene results in two transcript variants which encode the same isoform. [provided by RefSeq, Jul 2008]
Uniprot Description
BAD: a proapoptotic member of the Bcl-2 family. Displaces Bax from binding to Bcl-2 and Bcl-xL, resulting in cell death. Survival factors such as IL-3 can inhibit the apoptotic activity of Bad inducing the phosphorylation of Bad by Akt and p90RSK. 14-3-3 proteins bind phosphorylated Bad, inhibiting its binding to Bcl-2 and Bcl-xL. Phosphorylation by mitochondria-anchored PKA in the BH3 domain can block the dimerization of Bad and Bcl-xL.

Product Notes
The Bcl-xL bad (Catalog #AAA668541) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Polyclonal Antibody to Bcl-xL reacts with Dog, Gerbil, Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Bcl-xL can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC) Formalin/Paraffin, Immunoprecipitation (IP). WB: 1: 1000-1: 2000 IHC (paraffin): 1: 1000-1: 5000 IP: 1: 50-1: 200. Researchers should empirically determine the suitability of the Bcl-xL bad for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Bcl-xL, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
