Immunofluorescence (IF) (Fluorescent confocal image of HeLa cells stained with phospho-Bad-S99 antibody. HeLa cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with AP3039a phospho-Bad-S99 primary antibody (1:200, 2 h at room temperature). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 ug/ml, 5 min). Note the highly specific localization of the phospho-Bad-S99 mainly to the cytoplasm.)

Uniprot Description

BAD: a proapoptotic member of the Bcl-2 family. Displaces Bax from binding to Bcl-2 and Bcl-xL, resulting in cell death. Survival factors such as IL-3 can inhibit the apoptotic activity of Bad inducing the phosphorylation of Bad by Akt and p90RSK. 14-3-3 proteins bind phosphorylated Bad, inhibiting its binding to Bcl-2 and Bcl-xL. Phosphorylation by mitochondria-anchored PKA in the BH3 domain can block the dimerization of Bad and Bcl-xL.

Product Notes

The Bad bad (Catalog #AAA6464757) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Bad, Phosphorylated (Ser99) (Bcl2 Antagonist Of Cell Death, BAD, Bcl-2-binding Component 6, Bcl-XL/Bcl-2-Associated Death Promoter, Bcl-2-like 8 Protein, BAD, BBC6, BCL2L8) (APC) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Bad can be used in a range of immunoassay formats including, but not limited to, FLISA, Western Blot (WB), Dot Blot (DB), Immunohistochemistry (IHC). FLISA: 1:1,000 Dot Blot: 1:500 IHC: 1:50-1:100 WB: 1:100-1:500 Applications are based on unconjugated antibody. Researchers should empirically determine the suitability of the Bad bad for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Bad, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
