Application Notes
AP3B2 antibody can be used for detection of AP3B2 by Western blot at 1 - 2 mug/mL. Antibody can also be used for immunohistochemistry starting at 5 mug/mL. For immunofluorescence start at 20 mug/mL.

Preparation and Storage
AP3B2 antibody can be stored at 4 degree C for three months and -20 degree C, stable for up to one year. As with all antibodies care should be taken to avoid repeated freeze thaw cycles. Antibodies should not be exposed to prolonged high temperatures.

AP3B2 Antibody: The AP3 complex is a heterotetramer composed of two large adaptins (AP3D1, AP3B1 or AP3B2), a medium adaptin (AP3M1 or AP3M2) and a small adaptin (APS1 or AP3S2). The complex is associated with the Golgi region as well as more peripheral structures. It facilitates the budding of vesicles from the Golgi membrane and may be directly involved in trafficking to lysosomes. Unlike the homologous AP3B1, AP3B2-containing AP3 complexes are preferentially targeted to neuronal processes, and mice deficient in AP3B2 displayed compromised synaptic zinc stores.

NCBI Description

Adaptor protein-3 (AP3) is a heterotetrameric vesicle-coat protein complex. Some AP3 subunits are ubiquitously expressed, whereas others are expressed exclusively in neurons. The neuron-specific AP3 complex, which includes AP3B2, is thought to serve neuron-specific functions such as neurotransmitter release (Grabner et al., 2006 [PubMed 16788073]).[supplied by OMIM, Mar 2008]

AP3B2: Subunit of non-clathrin- and clathrin-associated adaptor protein complex 3 that plays a role in protein sorting in the late-Golgi/trans-Golgi network (TGN) and/or endosomes. The AP complexes mediate both the recruitment of clathrin to membranes and the recognition of sorting signals within the cytosolic tails of transmembrane cargo molecules. AP-3 appears to be involved in the sorting of a subset of transmembrane proteins targeted to lysosomes and lysosome-related organelles. Belongs to the adaptor complexes large subunit family. 2 isoforms of the human protein are produced by alternative splicing.
