Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot. Store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

AMP-activated protein kinase (AMPK) is highly conserved from yeast to plants and animals andplays a key role in the regulation of energy homeostasis 1). AMPK is a heterotrimeric complex composed of a catalytic a subunit and regulatory b and g subunits, each of which is encoded by two or three distinct genes a1, 2; b1, 2; g1, 2, 3) (2). The kinase is activated by an elevated AMP/ATP ratio due to cellular and environmental stress, such as heat shock, hypoxia and ischemia (1). Tumor suppressor LKB1, in association with accessory proteins STRAD and MO25, phosphorylates AMPK at Thr172 at the activation loop, and this phosphorylation is required for AMPK activation (3-5). AMPKa is also phosphorylated at Thr258 and Ser485 for a1; Ser491 for a2). The upstream kinase and the biological significance of these phosphorylation events have yet to be elucidated (6). The b1 subunit is posttranslationally modified by myristoylation and multi-site phosphorylation including Ser24/25, Ser96, Ser101, Ser108, and Ser182 (6,7). Phosphorylation at Ser108 of the b1 subunit seems to be required for the activation of AMPK enzyme, while phosphorylation at Ser24/25 and Ser182 affects AMPK localization (7). Several mutations in AMPKg subunits have been identified, most of which are located in the putative AMP/ATP binding sites (CBS or Bateman domains). Mutations at these sites lead to reduction of AMPK activity and cause glycogen accumulation in heart or skeletal muscle (1,2). Accumulating evidence indicates that AMPK not only regulates the metabolism of fatty acids and glycogen, but also modulates protein synthesis and cell growth through EF2 and TSC2/mTOR pathways, as well as blood flow via eNOS/nNOS (1).

Uniprot Description

Function: Regulatory subunit of the probable trimeric SNF1-related protein kinase (SnRK) complex, which may play a role in a signal transduction cascade regulating gene expression and carbohydrate metabolism in higher plants. The SnRK complex may also be involved in the regulation of fatty acid synthesis by phosphorylation of acetyl-CoA carboxylase and in assimilation of nitrogen by phosphorylating nitrate reductase.

Subunit structure: Subunit of a probable heterotrimeric complex consisting of an alpha catalytic (KIN10 or KIN11) subunit, and a beta (KINB) and a gamma (KING or SNF4) non-catalytic regulatory subunits. Interacts with SNF4. Ref.1 Ref.8 Ref.9

Tissue specificity: Expressed in leaves, stems, roots, flower buds and flowers. Not detectable in siliques. Ref.1

Induction: Repressed in the dark. Ref.1

Domain: Kinase-interacting sequence (KIS) is specific for the alpha catalytic subunit interaction and Association with SNF1 Complex (ASC) is specific for the gamma non-catalytic regulatory subunit interaction. Ref.7
