Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

AMPK is responsible for the regulation of fatty acid synthesis by phosphorylation of acetyl- CoA carboxylase. It also regulates cholesterol synthesis via phosphorylation and inactivation of hormone-sensitive lipase and hydroxymethylglutaryl-CoA reductase. It appears to act as a metabolic stress-sensing protein kinase switching off biosynthetic pathways when cellular ATP levels are depleted and when 5'-AMP rises in response to fuel limitation and/or hypoxia. AMPK alpha1, a member of the Ser/Thr protein kinase family, is a catalytic subunit of AMPK.

Uniprot Description

Function: Regulatory subunit of the probable trimeric SNF1-related protein kinase (SnRK) complex, which may play a role in a signal transduction cascade regulating gene expression and carbohydrate metabolism in higher plants. The SnRK complex may also be involved in the regulation of fatty acid synthesis by phosphorylation of acetyl-CoA carboxylase and in assimilation of nitrogen by phosphorylating nitrate reductase.

Subunit structure: Subunit of a probable heterotrimeric complex consisting of an alpha catalytic (KIN10 or KIN11) subunit, and a beta (KINB) and a gamma (KING or SNF4) non-catalytic regulatory subunits. Interacts with SNF4. Ref.1 Ref.8 Ref.9

Tissue specificity: Expressed in leaves, stems, roots, flower buds and flowers. Not detectable in siliques. Ref.1

Induction: Repressed in the dark. Ref.1

Domain: Kinase-interacting sequence (KIS) is specific for the alpha catalytic subunit interaction and Association with SNF1 Complex (ASC) is specific for the gamma non-catalytic regulatory subunit interaction. Ref.7
