NCBI Description

This gene encodes a mitochondrially localized enzyme that catalyzes the reversible formation of acetoacetyl-CoA from two molecules of acetyl-CoA. Defects in this gene are associated with 3-ketothiolase deficiency, an inborn error of isoleucine catabolism characterized by urinary excretion of 2-methyl-3-hydroxybutyric acid, 2-methylacetoacetic acid, tiglylglycine, and butanone. [provided by RefSeq, Feb 2009]

Uniprot Description

ACAT1: Plays a major role in ketone body metabolism. Defects in ACAT1 are a cause of 3-ketothiolase deficiency (3KTD); also known as alpha- methylacetoaceticaciduria. 3KTD is an inborn error of isoleucine catabolism characterized by intermittent ketoacidotic attacks associated with unconsciousness. Some patients die during an attack or are mentally retarded. Urinary excretion of 2-methyl-3- hydroxybutyric acid, 2-methylacetoacetic acid, triglylglycine, butanone is increased. It seems likely that the severity of this disease correlates better with the environmental or acquired factors than with the ACAT1 genotype. Belongs to the thiolase family.

Protein type: Amino Acid Metabolism - lysine degradation; Carbohydrate Metabolism - pyruvate; Mitochondrial; Secondary Metabolites Metabolism - terpenoid backbone biosynthesis; EC 2.3.1.9; Carbohydrate Metabolism - butanoate; Lipid Metabolism - synthesis and degradation of ketone bodies; Acetyltransferase; Carbohydrate Metabolism - propanoate; Lipid Metabolism - fatty acid; Amino Acid Metabolism - tryptophan; Amino Acid Metabolism - valine, leucine and isoleucine degradation

Chromosomal Location of Human Ortholog: 11q22.3

Cellular Component: mitochondrial inner membrane; mitochondrial matrix; mitochondrion

Molecular Function: acetyl-CoA C-acetyltransferase activity; coenzyme binding; enzyme binding; metal ion binding; protein homodimerization activity

Biological Process: brain development; branched chain family amino acid catabolic process; ketone body biosynthetic process; ketone body catabolic process; liver development; protein homooligomerization; response to hormone stimulus; response to organic cyclic substance; response to starvation

Disease: Alpha-methylacetoacetic Aciduria

Product Notes

The ACAT1 acat1 (Catalog #AAA421633) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The Goat anti-ACAT1 (aa253-266) Antibody reacts with Tested: Human, Mouse, Rat; Expected from sequence similarity: Human, Mouse, Rat, Dog, Cow and may cross-react with other species as described in the data sheet. AAA Biotech's ACAT1 can be used in a range of immunoassay formats including, but not limited to, Peptide ELISA (EIA), Western Blot (WB), Immunofluorescence (IF). Peptide ELISA: Antibody detection limit dilution 1: 2000. Western Blot: Approx 45kDa band observed in Human, Mouse and Rat Liver lysates (calculated MW of 45.2kDa according to Human NP_000010.1, 44.8kDa according to Mouse NP_659033.1 and 44.7kDa according to Rat NP_058771.2). Recommended concentration: 0.1-0.3ug/ml. Immunofluorescence: Customer finds particulate cytoplasm staining in HeLa. Researchers should empirically determine the suitability of the ACAT1 acat1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: DEEYKRVDFS KVPK. It is sometimes possible for the material contained within the vial of "ACAT1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
