Application Notes
Suitable for use in Direct ELISA, Western Blot. and Flow Cytometry.
Dilution: Direct ELISA: 0.5-1.0ug/ml.Detection limit for rmCD73 is
~0.3ng/well.
Western Blot: 0.1-0.2ug/ml. Detection limit for rmCD73 is ~1ng/lane under non-reducing and reducing conditions.
Flow Cytometry (Murine splenocytes): 25ug/ml. Add 100ul of the diluted solution to 1-2.5x10e5 cells in a total reaction volume not excedding 200ul.

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute by adding sterile 40-50% glycerol, PBS. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

CD73 (Ecto-5'nucleotidase, 5 '-NT) is a membrane-bound glycoprotein that functions to hydrolyze extracellular nucleoside monophosphates into bioactive nucleoside intermediates. This enables the uptake of adenosine, inosine, and guanosine into the cell and their subsequent reconversion into ATP and GTP in the purine salvage pathway. The expression of CD73 on neuronal cells has been described during developmental processes and, on lymphocytes, CD73 serves as a maturation marker, being absent from the surface of both immature B and T cells.

NCBI Description

The protein encoded by this gene is a plasma membrane protein that catalyzes the conversion of extracellular nucleotides to membrane-permeable nucleosides. The encoded protein is used as a determinant of lymphocyte differentiation. Defects in this gene can lead to the calcification of joints and arteries. Two transcript variants encoding different isoforms have been found for this gene.
