Preparation and Storage
May be stored at 4 degree C before opening. DO NOT FREEZE! Stable at 4 degree C as an undiluted liquid. Dilute only prior to immediate use. Stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Freezing APC conjugates will result in a substantial loss of activity. APC conjugates are sensitive to light.

CD73 (also known as [ecto]-5'-nucleotidase/5'-NT) is a variably glycosylated, 69-73kD member of the 5'-nucleotidase family of enzymes. It is expressed on multiple cell types, including vascular endothelium, transitional and nonkeratinized epithelium, cardiomyocytes, small intestine epithelium, FoxP3 Treg lymphocytes, FDCs, and B cells. CD73 hydrolyzes AMP to adenosine and phosphate. This creates diffusible nucleosides necessary for cell homeostasis, and a ligand for cell membrane adenosine receptors. Mature human CD73 is a 523aa GPI-linked protein (aa27-549). It contains a large Zn-dependent nucleotidase catalytic region (aa28-532) and a C-terminal substrate binding site (aa500-506). On the cell surface, it exists as a disulfide-linked homodimer. Two splice variants are reported. One shows a deletion of aa405-454, and a second possesses a 12aa substitution for aa253-574. Over aa1-511, human CD73 shares 88% aa identity with both mouse and rat CD73.

NCBI Description

The protein encoded by this gene is a plasma membrane protein that catalyzes the conversion of extracellular nucleotides to membrane-permeable nucleosides. The encoded protein is used as a determinant of lymphocyte differentiation. Defects in this gene can lead to the calcification of joints and arteries. Two transcript variants encoding different isoforms have been found for this gene.
