Principle of the Assay: NT4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-NT4 antibody and an NT4-HRP conjugate. The assay sample and buffer are incubated together with NT4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NT4 concentration since NT4 from samples and NT4-HRP conjugate compete for the anti-NT4 antibody binding site. Since the number of sites is limited, as more sites are occupied by NT4 from the sample, fewer sites are left to bind NT4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NT4 concentration in each sample is interpolated from this standard curve.

This gene is a member of a family of neurotrophic factors, neurotrophins, that control survival and differentiation of mammalian neurons. The expression of this gene is ubiquitous and less influenced by environmental signals. While knock-outs of other neurotrophins including nerve growth factor, brain-derived neurotrophic factor, and neurotrophin 3 prove lethal during early postnatal development, NTF5-deficient mice only show minor cellular deficits and develop normally to adulthood. NT-4/5: Target-derived survival factor for peripheral sensory sympathetic neurons. Defects in NTF4 may be associated with susceptibility to primary open angle glaucoma type 1O (GLC1O). A form of primary open angle glaucoma (POAG). POAG is characterized by a specific pattern of optic nerve and visual field defects. The angle of the anterior chamber of the eye is open, and usually the intraocular pressure is increased. The disease is asymptomatic until the late stages, by which time significant and irreversible optic nerve damage has already taken place. Belongs to the NGF-beta family.
