Principle of the Assay: The Human CD271 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human CD271 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Human CD271 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD271 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human CD271 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD271 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: Nerve growth factor receptor (NGFR) is also referred to as p75(NTR) because of its ability to bind at low affinity not only to NGF, but also other neurotrophins including brain-derived neurotrophic factor (BDNF), neurotrophin-3 and neurotrophin-4/5. The nerve growth factor receptor gene is at human chromosome region 17q12-17q22, distal to the chromosome 17 breakpoint in acute leukemias. The neurotrophin receptor p75(NTR), a tumor necrosis factor receptor superfamily member expressed in hepatic stellate cells after fibrotic and cirrhotic liver injury in humans, is a regulator of liver repair. In mice, depletion of p75(NTR) exacerbated liver pathology and inhibited hepatocyte proliferation in vivo. In addition, it is showed that neurotrophins activate p75(NTR) to induce apoptosis through the induction of the sphingomyelin (SM) cycle and increased production of ceramide. Overexpression of p75(NTR) is also found to activate the SM pathway.

NCBI Description

Nerve growth factor receptor contains an extracellular domain containing four 40-amino acid repeats with 6 cysteine residues at conserved positions followed by a serine/threonine-rich region, a single transmembrane domain, and a 155-amino acid cytoplasmic domain. The cysteine-rich region contains the nerve growth factor binding domain. [provided by RefSeq, Jul 2008]

Uniprot Description

NGFR: Low affinity receptor which can bind to NGF, BDNF, NT-3, and NT-4. Can mediate cell survival as well as cell death of neural cells. Homodimer; disulfide-linked. Interacts with p75NTR- associated cell death executor. Interacts with TRAF2, TRAF4, TRAF6, PTPN13 and RANBP9. Interacts through TRAF6 with SQSTM1 which bridges NGFR to NTRK1. Interacts with BEX1 and NGFRAP1/BEX3. Interacts with KIDINS220 and NTRK1. Can form a ternary complex with NTRK1 and KIDINS220 and this complex is affected by the expression levels of KIDINS220. An increase in KIDINS220 expression leads to a decreased association of NGFR and NTRK1. Interacts with NTRK2; may regulate the ligand specificity of the NTRK2 receptor. Interacts with LINGO1.

Product Notes

The Human NGFR ngfr (Catalog #AAA8248381) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8248381 ELISA Kit recognizes Human NGFR. It is sometimes possible for the material contained within the vial of "CD271, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
