Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 6 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Metabotropic glutamate receptors have been classified into three groups. In general, class I receptors (mGluR1 and mGluR5) mediate the excitation of neurons, whereas activation of class II (mGluR2 and mGluR3) and class III (mGluR4a, mGluR7a, mGluR7b and mGluR8) results in a depression of synaptic transmission. Class I mGluRs are localized in the postsynaptic membrane and are positively coupled to phospholipase C (PLC). Class II and class III mGluRs localize to the presynaptic membrane and are negatively coupled to adenylate cyclase. It has been speculated that mGluR2 and/or mGluR3 may play a role in the modulation of the mossy terminal function.

NCBI Description

L-glutamate is the major excitatory neurotransmitter in the central nervous system and activates both ionotropic and metabotropic glutamate receptors. Glutamatergic neurotransmission is involved in most aspects of normal brain function and can be perturbed in many neuropathologic conditions. The metabotropic glutamate receptors are a family of G protein-coupled receptors, that have been divided into 3 groups on the basis of sequence homology, putative signal transduction mechanisms, and pharmacologic properties. Group I includes GRM1 and GRM5 and these receptors have been shown to activate phospholipase C. Group II includes GRM2 and GRM3 while Group III includes GRM4, GRM6, GRM7 and GRM8. Group II and III receptors are linked to the inhibition of the cyclic AMP cascade but differ in their agonist selectivities. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jan 2009]
