Application Notes
Suitable for use in Western Blot and Immunocytochemistry.
Dilution: Western Blot: 0.1-0.5ug/ml. Analysis of rat brain shows bands migrating at Mr 100kD (which contains both mGluR2 and mGluR3) and 190kD. The larger band is thought to be a dimer of the smaller band.
Immunocytochemistry: 0.1-0.5ug/ml. Paraformaldehyde or paraformaldehyde/glutaraldehyde fixed tissue with light and electron microscopy. Cryostat and vibratome sections can be used with or without Triton X-100 treatment.

Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Metabotropic glutamate receptors have been classified into three groups. In general, class I receptors (mGluR1 and mGluR5) mediate the excitation of neurons, whereas activation of class II (mGluR2 and mGluR3) and class III (mGluR4a, mGluR7a, mGluR7b and mGluR8) results in a depression of synaptic transmission. Class I mGluRs are localized in the postsynaptic membrane and are positively coupled to phospholipase C (PLC). Class II and class III mGluRs are localized to the presynaptic membrane and are negatively coupled to adenylate cyclase. It has been speculated that mGluR2 and/or mGluR3 may play a role in the modulation of the mossy terminal function.
