Preparation and Storage

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Regulator Of G Protein Signaling 7 (RGS7). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Regulator Of G Protein Signaling 7 (RGS7). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Regulator Of G Protein Signaling 7 (RGS7), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Regulator Of G Protein Signaling 7 (RGS7) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

RGS7: inhibits signal transduction by increasing the GTPase activity of G protein alpha subunits thereby driving them into their inactive GDP-bound form. Activity on G(o)-alpha is specifically enhanced by the RGS6/GNG5 dimer. May play a role in synaptic vesicle exocytosis. May play important role in the rapid regulation of neuronal excitability and the cellular responses to short-lived stimulations. Heterodimer with GNG5. Interacts with RGS7BP, leading to regulate the subcellular location of the heterodimer formed with Gbeta5 (By similarity). Interacts with 14-3-3 protein Tau and SNAP25BP. Four alternatively spliced isoforms have been described.
