Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Related Product Information for GZMA elisa kit
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Granzyme A (GZMA). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Granzyme A (GZMA). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Granzyme A (GZMA), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Granzyme A (GZMA) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

Cytolytic T lymphocytes (CTL) and natural killer (NK) cells share the remarkable ability to recognize, bind, and lyse specific target cells. They are thought to protect their host by lysing cells bearing on their surface 'nonself' antigens, usually peptides or proteins resulting from infection by intracellular pathogens. The protein described here is a T cell- and natural killer cell-specific serine protease that may function as a common component necessary for lysis of target cells by cytotoxic T lymphocytes and natural killer cells. [provided by RefSeq, Jul 2008]

Uniprot Description

GZMA: This enzyme is necessary for target cell lysis in cell- mediated immune responses. It cleaves after Lys or Arg. Cleaves APEX1 after 'Lys-31' and destroys its oxidative repair activity. Involved in apoptosis. Homodimer; disulfide-linked. Interacts with APEX1. Dexamethasone (DEX) induces expression of isoform beta and represses expression of isoform alpha. The alteration in expression is mediated by binding of glucocorticoid receptor to independent promoters adjacent to the alternative first exons of isoform alpha and isoform beta. Belongs to the peptidase S1 family. Granzyme subfamily. 2 isoforms of the human protein are produced by alternative promoter.

Protein type: Secreted, signal peptide; Secreted; Protease; Apoptosis; EC 3.4.21.78

Chromosomal Location of Human Ortholog: 5q11-q12

Cellular Component: extracellular region; immunological synapse; nucleus

Molecular Function: protein binding; protein homodimerization activity; serine-type endopeptidase activity

Biological Process: proteolysis involved in cellular protein catabolic process; apoptosis; positive regulation of apoptosis; cytolysis; negative regulation of oxidoreductase activity; negative regulation of endodeoxyribonuclease activity; immune response; negative regulation of DNA binding

Product Notes

The Mouse GZMA gzma (Catalog #AAA2023967) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2023967 ELISA Kit recognizes Mouse GZMA. It is sometimes possible for the material contained within the vial of "Granzyme A (GZMA), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
