Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of EP300 in mouse tissue homogenates, cell lysates and other biological fluids.

Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to EP300. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to EP300. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain EP300, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of EP300 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

This gene encodes the adenovirus E1A-associated cellular p300 transcriptional co-activator protein. It functions as histone acetyltransferase that regulates transcription via chromatin remodeling and is important in the processes of cell proliferation and differentiation. It mediates cAMP-gene regulation by binding specifically to phosphorylated CREB protein. This gene has also been identified as a co-activator of HIF1A (hypoxia-inducible factor 1 alpha), and thus plays a role in the stimulation of hypoxia-induced genes such as VEGF. Defects in this gene are a cause of Rubinstein-Taybi syndrome and may also play a role in epithelial cancer. [provided by RefSeq, Jul 2008]

p300: a histone acetyltransferase and transcriptional co-activator that regulates transcription via chromatin remodeling. Acetylates all four core histones in nucleosomes. Related to CPB (CREB-binding protein), and like CPB can stimulate transcription through activation of CREB. Specifically inhibited by the adenovirus oncoprotein E1A. A co-activator of HIF1A (hypoxia-inducible factor 1 alpha), and thus plays a role in the stimulation of hypoxia-induced genes such as VEGF. Mediates cAMP-gene regulation by binding specifically to phosphorylated CREB protein. Methylated at R580 and R604 in the KIX domain by CARM1, which blocks association with CREB, inhibits CREB signaling and activates the apoptotic response. Also methylated at R2142 by CARM1, which impairs interaction with NCoA2.
