Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. An antibody specific for mouse CXCL12/SDF-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CXCL12/SDF-1 present is bound by the immobilized antibody. After washing away any unbound substances, and then a detection antibody specific for CXCL12/SDF-1 is added to the wells and binds to the combination of capture antibody CXCL12/SDF-1 in sample. Following a wash to remove any unbound combination, and enzyme conjugate is added to the wells. Following incubation and wash steps, a substrate solution is added to the wells and color develops in proportion to the amount of CXCL12/SDF-1 bound in the initial step. The color development is stopped and the absorbance is measured.
