Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Mouse AATK concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to AATK. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for AATK and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain AATK, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of AATK. You can calculate the concentration of AATK in the samples by comparing the OD of the samples to the standard curve.

NCBI Description

The protein encoded by this gene contains a tyrosine kinase domain at the N-terminus and a proline-rich domain at the C-terminus. This gene is induced during apoptosis, and expression of this gene may be a necessary pre-requisite for the induction of growth arrest and/or apoptosis of myeloid precursor cells. This gene has been shown to produce neuronal differentiation in a neuroblastoma cell line. Two transcript variants encoding different isoforms have been found for this gene. Lmr1: a protein kinase kinase of the Lmr family. Predominantly expressed in the nervous system and is involved in neurite extension and apoptosis of cerebellar granule cells. Contains a tyrosine kinase domain at the N-terminal end and a proline-rich domain at the C-terminal end. May be necessary for the induction of growth arrest and/or apoptosis of myeloid cells induced by cytokine withdrawal, such as IL3, and during G-CSF-induced terminal differentiation of myeloblasts to granulocytes. Four alternatively spliced isoforms have been reported.
