Specificity
Recognizes an epitope localized in the lectin domain of rat surfactant protein D (SP-D). Reacts with alveolar type II cells, alveolar macrophages, types of Clara cells SP-A+ and SP-A- and extracellular deposits. Does not react with Alveolar type III cells. No reaction on rat skin, colon, kidney or liver. Species Crossreactivity: human.

Application Notes
Suitable for use in Western Blot, Immunohistochemistry and ELISA.
Dilution: Immunohistochemistry (Use snap frozen or paraffin embedded tissue): Fresh rat tissue: 1:400; fresh human tissue: 1:20. Fixation: Acetone 5 minutes at 4 degree C, paraformaldehyde, paraffin. Tissue pretreatment: Microwave treatment 2 x 5 minutes at 750W for paraffin tissues.
ELISA: Use as a capture antibody.

Purified rat surfactant protein D from bronchoalveolar lavage.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Surfactant Protein D is a Ca2+ dependent carbohydrate binding protein with a structure similar to other C-type mammalian lectins. Its molecular weight is 620kD under non-reducing conditions and 43kD under reducing conditions. SP-D enhances the production of oxygen radicals in alveolar macrophages and regulates some actions of SP-A. SP-D is a lung surfactant associated collectin that has been implicated in the innate pulmonary host defense against a wide variety of important bacterial, fungal and viral pathogens.
