Application Notes
Suitable for use in Immunohistochemistry, Immunocytochemistry and Western Blot.
Dilution: Immunohistochemistry (Acetone fixed cryostat sections): 0.5-10ug/ml using an APAAP or PAP procedure. Antibody also reacts with 4% PFA fixed frozen tissue. Typical block contains 1% non-fat dry milk or 3% NGS and 0.2-1.5% triton X-100 (higher triton is only used on thick sections; blocked for 1-3 hours at RT. Primary antibody is diluted only in PBS or TBS without excess blocking materials (no triton or milk); typical dilution is 1:200-1:1000 with incubations up to 48 hours at 4 degree C. Wash 3x10' in PBS and detect via DAB. Immunoreactive neurons are characterized by the dark brown, fine granular deposition in soma and proximal dendrites.
Immunocytochemistry (cell smears): 0.5-10ug/ml.
Western Blot: Under reducing conditions a band at ~25kD will be identified.

Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Superoxide dismutases (SOD) are found in all aerobic organisms. Their physiological function appears to be to protect cells from free radicals by scavenging superoxide anions. They catalyse the dismutation of superoxide anions to oxygen and hydrogen peroxide. SOD(Mn) is located primarily in mitrochondria and is thought to be of prokaryote origin. SOD(Mn) is a tetramer of identical subunits of MW 25kD by SDS-PAGE.

Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
