Western Blot (WB)

(Detection of Human S100A2 by Western Blot. Western blot shows lysates of A431 human epithelial carcinoma cell line. PVDF Membrane was probed with 2ug/ml of Human S100A2 Monoclonal Antibody followed by HRP­conjugated Mouse IgG Secondary Antibody. A specific band was detected for S100A2 at approximately 10kD (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.)

S100A2 is a 10kD member of the S100 family, EF-hand superfamily of Ca-binding proteins. It is expressed by multiple cell types, including keratinocytes, chondrocytes, and bronchial epithelium. S100A2 regulates both Ca and Zn (in human) within cells, and increases p53 activity. It forms both noncovalent and covalent homodimers, and a homotetramer under certain conditions. Human S100A2 is 98aa in length. It contains two EF-hand motifs aa13-48, 51-86 plus a calcium aa64-75 and zinc aa17-22 binding site. There is one potential alternative splice form that shows a 40aa insertion after Gly48. Full-length human S100A2 shares 89%, 85% aa identity with bovine and canine S100A2.

Uniprot Description

Function: May function as calcium sensor and modulator, contributing to cellular calcium signaling. May function by interacting with other proteins, such as TPR-containing proteins, and indirectly play a role in many physiological processes. May also play a role in suppressing tumor cell growth. Ref.1 Ref.8

Subunit structure: Homodimer. Interacts with FKBP4. Interacts with PPP5C (via TPR repeats); the interaction is calcium-dependent and modulates PPP5C activity. Ref.6 Ref.8 Ref.9

Tissue specificity: A subset of epithelial cells including normal human mammary epithelial cells and keratinocytes. Ref.1

Developmental stage: Preferentially expressed in normal human mammary epithelial cells as opposed to tumor-derived ones. The level of S100L was shown to correlate inversely with tumor progression. Ref.2

Induction: By growth factors in early G1 phase and probably by cell-cycle regulation in S phase. DNA methylation probably plays a direct negative role in suppressing S100L gene expression in tumor cells. Ref.1 Ref.2
