Related Product Information for anti-PDK1 antibody
PDK1 (pyruvate dehydrogenase kinase isoform 1) is involved in the regulation of enzymatic activity of mammalian pyruvate dehydrogenase (PDH) that is a part of a mitochondrial multienzyme complex to catalyze the oxidative decarboxylation of pyruvate and is one of the major enzymes responsible for the regulation of homeostasis of carbohydrate fuels in mammals. PDK1 has been found to serve as an effective therapeutic target for inhibition of glioblastoma growth.

NCBI Description

Encodes 3-phosphoinositide-dependent protein kinase that contains pleckstrin homology domain and binds 3-phosphoinositides. It activates the protein kinase AGC2-1 in a phosphatidic acid dependent manner. Phosphorylates S6K1. Interacts with PID, and transphosphorylation by PDK1 increases PID autophosphorylation.

Uniprot Description

Function: May couple lipid signals to the activation-loop phosphorylation of several protein kinases of the so-called AGC kinase family. Interacts via its pleckstrin homology domain with phosphatidic acid, PtdIns3P and PtdIns(3,4)P2 and to a lesser extent with PtdIns(4,5)P2 and PtdIns4P. May play a general role in signaling processes controlling the pathogen/stress response, polar auxin transport and development. Transphosphorylates the AGC protein kinases OXI1/AGC2-1, PK1/S6K1, PK19/S6K2 and PID resulting in their activation. Ref.1 Ref.7 Ref.9 Ref.11 Ref.12

Catalytic activity: ATP + a protein = ADP + a phosphoprotein. Ref.6

Enzyme regulation: Activated by phosphatidic acid (PA) and in response to the fungal elicitor xylanase. Ref.10

Subunit structure: Interacts with the C-terminal PIF domain of the protein kinases D6PK/AGC1-1, OXI1/AGC2-1 and PID. Ref.7 Ref.11

Subcellular location: Cytoplasm

By similarity. Membrane; Peripheral membrane protein

By similarity. Note: Membrane-associated after cell stimulation

Tissue specificity: Ubiquitous. Ref.7

Domain: The PH domain is responsible for the interaction with the 3-phosphoinositides. The activation loop within the kinase domain is the target of phosphorylation. The PIF-binding region in the kinase domain of PDK1 acts as a docking site, enabling it to interact with and enhance the phosphorylation of substrates containing the PIF motif.

Post-translational modification: Phosphorylation on Thr-211 in the activation loop is required for full activity. PDK1 itself can autophosphorylate Thr-211, leading to its own activation.

Sequence similarities: Belongs to the protein kinase superfamily. AGC Ser/Thr protein kinase family. PDK1 subfamily.Contains 1 PH domain.Contains 1 protein kinase domain.

Sequence caution: The sequence CAB85557.1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally extended.

Product Notes

The PDK1 pdk1 (Catalog #AAA605315) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The PDK1 (PDPK1, PI-P3 Dependent Kinase, 3a Phosphoinositide dependent Protein Kinase 1) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's PDK1 can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB). Suitable for use in ELISA, Western Blotting. Dilution: Western Blotting: 1:250-1:1000. Researchers should empirically determine the suitability of the PDK1 pdk1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PDK1, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
