Application Notes
Suitable for use in Immunofluorescence, Western Blot, Immunohistochemistry and Immunoprecipitation.
Dilution: Western Blot: 0.5-1ug/ml for 2hrs at RT.
Immunohistochemistry (Formalin/paraffin): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 minutes followed by cooling at RT for 20 minutes.
Immunoprecipitation (Native and denatured): 2ug/mg protein lysate. Use Protein G.

Delipidated human milk fat globule membrane preparation. Cellular Localization: Cell membrane and cytoplasmic.
Hybridoma
P3-X63-Ag8.653 myeloma cells with spleen cells from BALB/c mice.
Positive Control
SKBR-3 cells. Breast, colon, or lung tissues.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

HMFG1 is considered as a differentiation marker. Uniprot Description

MFGE8: Plays an important role in the maintenance of intestinal epithelial homeostasis and the promotion of mucosal healing. Promotes VEGF-dependent neovascularization. Contributes to phagocytic removal of apoptotic cells in many tissues. Specific ligand for the alpha-v/beta-3 and alpha-v/beta-5 receptors. Also binds to phosphatidylserine-enriched cell surfaces in a receptor-independent manner. Zona pellucida-binding protein which may play a role in gamete interaction. Binds specifically to rotavirus and inhibits its replication. 3 isoforms of the human protein are produced by alternative splicing.

Research Articles on Mfge8

1. MFG-E8 attenuates inflammation and ALI after gut I/R and may represent a novel therapeutic agent.
2. Mfge8 directly bound collagen, and Mfge8-/- macrophages exhibited defective collagen uptake that could be rescued by recombinant Mfge8 containing at least one discoidin domain.
3. Data suggest that milk-fat globule-EGF factor 8 seems to act on the phagocytosis of apoptotic granulosa cells via macrophages and contribute to the regression process of atretic follicles.
4. Recombinant murine MFGE8 is able to reconstitute the clearance of apoptotic cells in septic rats to levels observed in sham operated animals.
