Applicable Applications for anti-MHC Class I antibody
Flow Cytometry (FC/FACS), ELISA (EIA)
Performance
Cultured human SW480 cells were harvested by EDTA. Five x 105 cells per tube were washed and pre incubated ~5 minutes with 20 ul human IgG at 250 ug/ml (to block non specific binding) after which they were incubated 45 minutes on ice with 80 ul of anti-human CD326/Biotin at concentration of 5 ug/ml. They were then washed twice and incubated with 2 degree reagent Streptavidin/R-PE after which they were washed twice, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.93 log10 fluorescent units when compared to an Isotype control Mouse IgG1/Biotin. Binding was blocked when cells were pre incubated with unlabeled anti-human CD326 mAb.
Conjugation
Biotin
Production
Antibody from (low FBS containing) tissue culture supernatant was Protein A purified to >95% mouse immunoglobulin by SDS-PAGE (<1% bovine immunoglobulin), and reacted with NHS-Biotin. Unconjugated Biotin was removed from conjugate using a desalting column.
Buffer
50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 5% Glycerol, 0.2% BSA, 0.04% NaN3 (as a preservative).
Immunogen
Recombinant Human CD326
Preparation and Storage
Store at 2 - 5 degree C. Freeze/thawing not recommended.
Product should retain activity for at least 12 months after shipping date when stored as recommended.

Related Product Information for anti-MHC Class I antibody
Information: Human CD326 (EpCAM, EGP2, HEA) is a 40kD glycoprotein expressed on epithelial cells and many carcinomas. It is an adhesion molecule whose function is implicated in carcinogenesis and erythropoesis. Antibody ANC8D4 recognizes recombinant CD326 in EIA, and cell surface human CD326 in FACS.

Product Notes

The MHC Class I (Catalog #AAA666614) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The anti-human MHC Class I-Biotin reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's MHC Class I can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), ELISA (EIA). Researchers should empirically determine the suitability of the MHC Class I for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MHC Class I, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
