Application Notes
Suitable for use in ELISA and Flow Cytometry.
Dilution: ELISA (Capture): 2-8ug/m. Can be used as a capture reagent in a sandwich immunoassay in combination with a biotinylated, mouse IL-17E antibody and recombinant mouse IL-17E, as the standard.
Flow Cytometry: For intracellular flow cytometry to detect IL-17E, cells must first be fixed and permeabilized with 4% paraformaldehyde and 0.1% saponin in PBS. Dilute this antibody to 25ug/ml and add 10ul of the diluted solution to 1-5 x 10e5 cells in a total reaction volume not exceeding 200ul. The binding of unlabeled antibody may be visualized by adding a secondary developing reagent such as anti-rat IgG conjugated to a fluorochrome.

Produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a rat immunized with purified, E. coli-derived, recombinant mouse IL-17E (rmIL-17E; aa 17-169; Accession # NP_542767).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C for short-term only. Reconstitute with sterile 40-50% glycerol, PBS. Aliquot and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

NCBI Description

The protein encoded by this gene is a cytokine that shares sequence similarity with interleukin 17. This cytokine can induce NF-kappaB activation, and stimulate the production of interleukin 8. Both this cytokine and interleukin 17B are ligands for the cytokine receptor IL17BR. Studies of a similar gene in mice suggest that this cytokine may be a pro-inflammatory cytokine favoring the Th2-type immune response. Alternative splicing results in multiple transcript variants. Researchers should empirically determine the suitability of the IL25 il25 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Interleukin 17E, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening.
