Principle of the Assay: IL-25 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-IL-25 antibody and an IL-25-HRP conjugate. The assay sample and buffer are incubated together with IL-25-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-25 concentration since IL-25 from samples and IL-25-HRP conjugate compete for the anti-IL-25 antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-25 from the sample, fewer sites are left to bind IL-25-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-25 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a cytokine that shares sequence similarity with interleukin 17. This cytokine can induce NF-kappaB activation, and stimulate the production of interleukin 8. Both this cytokine and interleukin 17B are ligands for the cytokine receptor IL17BR. Studies of a similar gene in mice suggest that this cytokine may be a pro-inflammatory cytokine favoring the Th2-type immune response. Alternative splicing results in multiple transcript variants.
