Mouse anti-Human, Hsp90 Monoclonal Antibody | anti-Hsp90 antibody
Hsp90 antibody
Western Blot (WB)
(The Recombinant Human HSP90 (50ng) and Cell lysates(40ug) were resolved by SDS-PAGE, transferred to PVDF membrane and probed with anti-human HSP90antibody (1:1000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and an ECL detection systemLane 1. : Hsp90 recombinant protein Lane 2. : 293T cell lysate Lane 3. : HeLa cell lysate Lane 4. : HepG2 cell lysate)
Flow Cytometry (FC)
(Flow cytometry analysis of HSP90in HepG2cell line, staining at 2-5ug for 1x106cellsredline). The secondary antibody used goat anti-mouse IgGAlexa fluor 488 conjugate. Isotype control antibody was mouse IgG(black line).)
Immunohistochemistry (IHC)
(Paraffin embedded sections of human urothelium were incubated with anti-human Hsp90 (1:100)for 2 hours at room temperature. Antigen retrieval was performed in 0.1M sodium citrate buffer and detectedusing Diaminobenzidine (DAB))
Testing Data(TD)
(ICC/IF analysis of HSP90in HeLa cells line, stained with DAPI(Blue) for nucleus staining and monoclonal anti-human HSP90 antibody (1: 100) with goat anti-mouse IgG-Alexa fluor 488 conjugate(Green).)
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is an inducible molecular chaperone that functions as a homodimer. The encoded protein aids in the proper folding of specific target proteins by use of an ATPase activity that is modulated by co-chaperones. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jan 2012]
Uniprot Description
Molecular chaperone that promotes the maturation, structural maintenance and proper regulation of specific target proteins involved for instance in cell cycle control and signal transduction. Undergoes a functional cycle that is linked to its ATPase activity which is essential for its chaperone activity. This cycle probably induces conformational changes in the client proteins, thereby causing their activation. Interacts dynamically with various co-chaperones that modulate its substrate recognition, ATPase cycle and chaperone function (PubMed:11274138, PubMed:15577939, PubMed:15937123, PubMed:27353360). Engages with a range of client protein classes via its interaction with various co-chaperone proteins or complexes, that act as adapters, simultaneously able to interact with the specific client and the central chaperone itself. Recruitment of ATP and co-chaperone followed by client protein forms a functional chaperone. After the completion of the chaperoning process, properly folded client protein and co-chaperone leave HSP90 in an ADP-bound partially open conformation and finally, ADP is released from HSP90 which acquires an open conformation for the next cycle (PubMed:27295069, PubMed:26991466). Apart from its chaperone activity, it also plays a role in the regulation of the transcription machinery. HSP90 and its co-chaperones modulate transcription at least at three different levels. In the first place, they alter the steady-state levels of certain transcription factors in response to various physiological cues. Second, they modulate the activity of certain epigenetic modifiers, such as histone deacetylases or DNA methyl transferases, and thereby respond to the change in the environment. Third, they participate in the eviction of histones from the promoter region of certain genes and thereby turn on gene expression (PubMed:25973397). Binds bacterial lipopolysaccharide (LPS) and mediates LPS-induced inflammatory response, including TNF secretion by monocytes (PubMed:11276205). Antagonizes STUB1-mediated inhibition of TGF-beta signaling via inhibition of STUB1-mediated SMAD3 ubiquitination and degradation (PubMed:24613385).

Product Notes
The Hsp90 hsp90aa1 (Catalog #AAA200047) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Hsp90 antibody reacts with Human, and may cross-react with other species as described in the data sheet. AAA Biotech's Hsp90 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), WB (Cell lysate), Immunohistochemistry (IHC). The antibody has been tested by ELISA, Western Blot, and Immunohistochemistry analysis to assure specifity and reactivity. Since application varies, however, each investigation should be titrated by the reagent to obtain optimal results. Recommended diltuion range for Western Blot is 1:500~ 1:200 and immunohistochemistry analysis is 1:100~200. Recommended starting dilution for Western Blot is 1:1000 and Immunohistochemistry is 1:100. Researchers should empirically determine the suitability of the Hsp90 hsp90aa1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Hsp90, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
