NCBI Description

This gene was identified by the reactivity of its encoded protein to a monoclonal antibody prepared against brain homogenates from patients with Alzheimer's disease. Analysis of the original protein (fetal Alz-50 reactive clone 1, or FAC1), identified as an 810 aa protein containing a DNA-binding domain and a zinc finger motif, suggested it might play a role in the regulation of transcription. High levels of FAC1 were detected in fetal brain and in patients with neurodegenerative diseases. The protein encoded by this gene is actually much larger than originally thought, and it also contains a C-terminal bromodomain characteristic of proteins that regulate transcription during proliferation. The encoded protein is highly similar to the largest subunit of the Drosophila NURF (nucleosome remodeling factor) complex. In Drosophila, the NURF complex, which catalyzes nucleosome sliding on DNA and interacts with sequence-specific transcription factors, is necessary for the chromatin remodeling required for transcription. Two alternative transcripts encoding different isoforms have been described completely. [provided by RefSeq, Jul 2008]

Uniprot Description

FALZ: Histone-binding component of NURF (nucleosome-remodeling factor), a complex which catalyzes ATP-dependent nucleosome sliding and facilitates transcription of chromatin. Specifically recognizes H3 tails trimethylated on 'Lys-4' (H3K4me3), which mark transcription start sites of virtually all active genes. May also regulate transcription through direct binding to DNA or transcription factors. Belongs to the PBTF family. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: Transcription factor; DNA-binding

Chromosomal Location of Human Ortholog: 17q24.3

Cellular Component: nucleoplasm; cytoplasm; nucleus

Molecular Function: DNA-dependent ATPase activity; protein binding; zinc ion binding; sequence-specific DNA binding; transcription factor binding

Biological Process: anterior/posterior pattern formation; chromatin remodeling; embryonic placenta development; regulation of transcription, DNA-dependent; transcription, DNA-dependent; positive regulation of transcription, DNA-dependent; brain development; negative regulation of transcription from RNA polymerase II promoter; endoderm development

Product Notes

The FAC1 bptf (Catalog #AAA249006) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Mouse Monoclonal [clone 2F10] (IgG2b) to Human FAC1 / BPTF reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's FAC1 / BPTF can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC - Paraffin), Western Blot (WB), ELISA (EIA). ELISA (1:10000), IHC-P (1:50), WB (1:500 - 1:2000). Researchers should empirically determine the suitability of the FAC1 bptf for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "FAC1 / BPTF, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
