Specificity
Recognizes all known forms of the Mr 74kD intermediate chain subunit (IC74) of mammaliam cytoplasmic dynein. The antigenic site has been localized to the first 60 amino acids of the amino terminal portion of the molecule (a region conserved in both IC74 genes). Species Crossreactivity: Rat, mouse, bovine, sheep, fish, Drosophila and Xenopus. Does not react with squid.

Application Notes
Suitable for use in Immunocytochemistry, Immunoblot and Immunoprecipitation.
Dilution: Immunocytochemistry: 1:50-1:100. Reacts with cultured MDCK cells, NRR cells, N1E cells and PTK-1 cells.
Immunoprecipitation: 10ul/0.5g tissue. The monoclonal clearly immunoprecipitates the entire dynein complex stoichiometrically from TX-100 or NP-40 lysates (including the 530kD heavy chain, the light intermediate chains and the light chains) from various tissues and cultured cell lines. If detergents such as SDS are used to prepare the lysates the only dynein subunits which are immunoprecipitated are the IC74 subunits (presumably because the dynein complex dissociates), and other contaminating proteins can be found in SDS immunoprecipitates.

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Researchers should empirically determine the suitability of the Dynein, Cytoplasmic, 74kD Intermediate Chains for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Dynein, Cytoplasmic, 74kD Intermediate Chains, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
