Related Product Information for anti-DNMT3L antibody
Methylation of DNA at cytosine residues plays an important role in regulation of gene expression, genomic imprinting and is essential for mammalian development. Hypermethylation of CpG islands in tumor suppressor genes or hypomethylation of bulk genomic DNA may be linked with development of cancer. To date, 3 families of mammalian DNA methyltransferase genes have been identified which include Dnmt1, Dnmt2 and Dnmt3. Dnmt1 is constitutively expressed in proliferating cells and inactivation of this gene causes global demethylation of genomic DNA and embryonic lethality. Dnmt2 is expressed at low levels in adult tissues and its inactivation does not affect DNA methylation or maintenance of methylation. The Dnmt3 family members, Dnmt3a and Dnmt3b, are strongly expressed in ES cells but their expression is down regulated in differentiating ES cells and is low in adult somatic tissue (1-6). Studies show that DNMT3L regulates the activity of DNMT3A and DNMT3B and stimulates their catalytic activities (7). DNMT3L has specifically been linked to the process of carcinogenesis, thru its role in nuclear

NCBI Description

CpG methylation is an epigenetic modification that is important for embryonic development, imprinting, and X-chromosome inactivation. Studies in mice have demonstrated that DNA methylation is required for mammalian development. This gene encodes a nuclear protein with similarity to DNA methyltransferases. This protein is not thought to function as a DNA methyltransferase as it does not contain the amino acid residues necessary for methyltransferase activity. However, this protein does stimulate de novo methylation by DNA cytosine methyltransferase 3 alpha and it is thought to be required for the establishment of maternal genomic imprints. This protein also mediates transcriptional repression through interaction with histone deacetylase 1. Alternative splicing results in two transcript variants. An additional splice variant has been described but its biological validity has not been determined. [provided by RefSeq]

Uniprot Description

DNMT3L: Catalytically inactive regulatory factor of DNA methyltransferases. It is essential for the function of DNMT3A and DNMT3B. Activates DNMT3A and DNMT3B by binding to their catalytic domain. Accelerates the binding of DNA and AdoMet to the methyltransferases and dissociates from the complex after DNA binding to the methyltransferases. Recognizes unmethylated histone H3 lysine 4 (H3K4) and induces de novo DNA methylation by recruitment or activation of DNMT3. Belongs to the C5-methyltransferase family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The DNMT3L dnmt3l (Catalog #AAA608699) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The DNMT3L (DNA (cytosine-5-)-methyltransferase 3-like MGC1090) reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's DNMT3L can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunoprecipitation (IP), Immunocytochemistry (ICC). Dilution: Western Blot:1-10ug/ml Immunocytochemistry:0.1-1ug/ml. Researchers should empirically determine the suitability of the DNMT3L dnmt3l for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "DNMT3L, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
