Short Description: Full-length recombinant DNMT3L protein (accession number NP_787063.1) was expressed in Sf9 cells and contains an N-terminal 6xHis-tag with a molecular weight of 46.6 kDa. The recombinant protein is >80% pure by SDS-PAGE. It is suitable for use in the study of enzyme kinetics, inhibitor screening, and selectivity profiling.

Background: DNMT3L (DNA (Cytosine-5-)-Methyltransferase 3-Like) is a nuclear protein with similarity to the DNMT family of proteins, but does not function as a DNA methyltransferase. It serves as an essential regulator of DNMT3A and DNMT3B, recruiting them to sites for deposition of de novo DNA methylation. DNMT3L is expressed in the presumptive gametes in mammalian embryos and is required for the establishment of genomic imprinting. DNMT3L has a PHD-like domain that allows binding to chromatin through interactions with the amino terminal tail of histone H3 that is devoid of lysine 4 methylation.

NCBI Description

CpG methylation is an epigenetic modification that is important for embryonic development, imprinting, and X-chromosome inactivation. Studies in mice have demonstrated that DNA methylation is required for mammalian development. This gene encodes a nuclear protein with similarity to DNA methyltransferases. This protein is not thought to function as a DNA methyltransferase as it does not contain the amino acid residues necessary for methyltransferase activity. However, this protein does stimulate de novo methylation by DNA cytosine methyltransferase 3 alpha and it is thought to be required for the establishment of maternal genomic imprints. This protein also mediates transcriptional repression through interaction with histone deacetylase 1. Alternative splicing results in two transcript variants. An additional splice variant has been described but its biological validity has not been determined. [provided by RefSeq]
