Dilution: Neutralization (ND50): ~0.3-1.5ug/ml in the presence of 7ng/ml of recombinant human CXCL11/l-TAC, using the human CXCR3 transfected BaF/3 cells in a chemotaxis assay.
The exact concentration required to neutralize human cell surface CXCR3 activity is dependent on the ligand concentration as well as the number of CXCR3 receptors present on the cell surface.
Flow Cytometry: 2.5x10e6 cells using whole human blood lymphocytes.
Immunohistochemistry: 8-25ug/ml detected CXCR3 in immersion fixed paraffin-embedded sections of human tonsil.
Testing Data
(Recombinant Human CXCL11/I‑TAC chemoattracts the BaF3 mouse pro‑B cell line transfected with human CXCR3 in a dose-dependent manner (orange line). The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin. Chemotaxis elicited by Recombinant Human CXCL11/I‑TAC (7ng/ml) is neutralized (green line) by increasing concentrations of MBS600325. The ND50 is typically 0.3-1.5µg/ml.)

This gene encodes a G protein-coupled receptor with selectivity for three chemokines, termed CXCL9/Mig (monokine induced by interferon-g), CXCL10/IP10 (interferon-g-inducible 10 kDa protein) and CXCL11/I-TAC (interferon-inducible T cell a-chemoattractant). Binding of chemokines to this protein induces cellular responses that are involved in leukocyte traffic, most notably integrin activation, cytoskeletal changes and chemotactic migration. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. One of the isoforms (CXCR3-B) shows high affinity binding to chemokine, CXCL4/PF4 (PMID:12782716).
