Application Notes
Suitable for use in Neutralization, Flow Cytometry and Immunohistochemistry.
Dilution: Neutralization (ND50): ~0.3-1.5ug/ml in the presence of 7ng/ml of recombinant human CXCL11/l-TAC, using the human CXCR3 transfected BaF/3 cells in a chemotaxis assay.
The exact concentration required to neutralize human cell surface CXCR3 activity is dependent on the ligand concentration as well as the number of CXCR3 receptors present on the cell surface.
Flow Cytometry: 2.5x10e6 cells using whole human blood lymphocytes.
Immunohistochemistry: 8-25ug/ml detected CXCR3 in immersion fixed paraffin-embedded sections of human tonsil.

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

NCBI Description

This gene encodes a G protein-coupled receptor with selectivity for three chemokines, termed CXCL9/Mig (monokine induced by interferon-g), CXCL10/IP10 (interferon-g-inducible 10 kDa protein) and CXCL11/I-TAC (interferon-inducible T cell a-chemoattractant). Binding of chemokines to this protein induces cellular responses that are involved in leukocyte traffic, most notably integrin activation, cytoskeletal changes and chemotactic migration. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. One of the isoforms (CXCR3-B) shows high affinity binding to chemokine, CXCL4/PF4 (PMID:12782716).
