• Performance: Five x 105 ficoll prepared human PBL were incubated 45 minutes on ice with 80 ul of anti-CD60b/FITC at a 1:50 dilution factor (10 ug /ml). Cells were then washed three times, fixed and analyzed by FACS. A 39% sub population of the cells stained positive with a mean shift of 2.3 log10 fluorescent units when compared to a Mouse IgM/FITC negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20 ul of 0.5 mg/ml anti-CD60b antibody.

• Production: Antibody from (low FBS containing) tissue culture supernatant was size exclusion purified to 90% Immunoglobulin by SDS-PAGE, and reacted with FITC. Unconjugated FITC was removed from conjugate using a desalting column. Antibody conjugate is at 0.5 mg/ml with a Fluorescein/IgM molar ratio of 68:1.
• Buffer: 50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 5% Glycerol, 0.2% BSA, 0.04% NaN3 (as a preservative).
• Immunogen: Human rheumatoid synovial T cell line (ST-1)
• Preparation and Storage: Store at 2 - 5 degree C. Freeze/Thawing is not recommended. Protect from light; Product should retain activity for at least 12 months after shipping date when stored as recommended.

Information: Human CD60 antigen is a glycolipid determinant found on about 30% of peripheral blood T cells. The glycolipid determinant is linked to a glycoprotein which can transduce activation signals. Antibody UM4D4 recognizes a 9-0-acetylated disialosyl group linked to the glycoprotein CD60. Antibody UM4D4 will activate T cells.
