• Application Notes: 1:50 (or use 1.6ul of concentrated stock per 5 x 105-cell test)
• Performance: Five x 105 cultured Nalm-6 cells were washed and incubated 45 minutes on ice with 80 ul of anti-CD58/FITC at a 1:50 dilution factor (10 ug/ml). Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 0.6 log10 fluorescent units when compared to a Mouse IgG1/FITC negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20 ul of 0.5 mg/ml anti-CD58 antibody.

• Production: Protein A purified antibody from tissue culture supernatant was reacted with FITC. Unconjugated FITC was separated from antibody/FITC conjugate by desalting column. The antibody/FITC conjugate is at 0.5 mg/ml with a Fluorescein/IgG molar ratio of 5.5.
• Buffer: 50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 5% Glycerol, 0.2% BSA, 0.04% NaN3 (as a preservative).

Information: Human CD58 is a cell surface glycoprotein that functions as an adhesion molocule and mediates costimulatory activity through its ligand CD2, which is expressed on T and natural killer cells (1,2). CD58 has wide cellular distribution on both hemopoietic and non-hemopoietic cells with strong expression on macrophages and memory T cells. Antibody TS2 recognizes the CD58 glycoprotein of about 60-70 kd and will inhibit HLA-DR mediated T cell cytotoxicity.

This gene encodes a member of the immunoglobulin superfamily. The encoded protein is a ligand of the T lymphocyte CD2 protein, and functions in adhesion and activation of T lymphocytes. CD58: Ligand of the T-lymphocyte CD2 glycoprotein. This interaction is important in mediating thymocyte interactions with thymic epithelial cells, antigen-independent and -dependent interactions of T-lymphocytes with target cells and antigen- presenting cells and the T-lymphocyte rosetting with erythrocytes. In addition, the LFA-3/CD2 interaction may prime response by both the CD2+ and LFA-3+ cells. 3 isoforms of the human protein are produced by alternative splicing.
