Performance
Five x 105 cultured Raji cells were washed and incubated 45 minutes on ice with 80 ul of anti-CD54(8.4A6)/Biotin at a concentration of 5 ug/ml. Cells were then washed twice and incubated 45 minutes with 2 degree reagent Streptavidin/R-Phycoerythrin, after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 2.66 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20 ul of 0.5 mg/ml anti-CD54(8.4A6) antibody.

Antibody from (low FBS containing) tissue culture supernatant was Protein A purified to >95% mouse immunoglobulin by SDS-PAGE (<1% bovine immunoglobulin), and reacted with NHS-Biotin. Unconjugated Biotin was removed from conjugate using a desalting column.

Information: Human CD54 (ICAM-1) mediates cell adhesion by binding to the integrins CD11a/CD18 (LFA-1) and to CD11b/CD18 (Mac-1). CD54 expression on resting peripheral blood leukocytes is weak but is upregulated on activated T and B lymphocytes and monocytes. Antibody 8.4A6 recognizes the D2 domain of the CD54 molecule of 90 kd. Antibody 8.4A6 inhibits CD54 binding to LFA-1.
