Flow Cytometry (FC/FACS)
(Flow cytometry using the anti-CD40 antibody chi220 (MBS488792). Daudi cells were fixed using 2% PFA and stained with anti-unknown specificity antibody (MBS488149; isotype control, black line) or the rabbit IgG1 version of chi220 (, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.)
This antibody was generated by immunizing female BALB/c mice with a fusion protein containing extracellular domain of human CD40 fused to the hinge CH2 and CH3 domains of a murine IgG2b antibody.

The protein encoded by this gene is expressed on the surface of T cells. It regulates B cell function by engaging CD40 on the B cell surface. A defect in this gene results in an inability to undergo immunoglobulin class switch and is associated with hyper-IgM syndrome. [provided by RefSeq, Jul 2008]

CD40LG: Mediates B-cell proliferation in the absence of co- stimulus as well as IgE production in the presence of IL-4. Involved in immunoglobulin class switching. Defects in CD40LG are the cause of X-linked immunodeficiency with hyper-IgM type 1 (HIGM1); also known as X-linked hyper IgM syndrome (XHIM). HIGM1 is an immunoglobulin isotype switch defect characterized by elevated concentrations of serum IgM and decreased amounts of all other isotypes. Affected males present at an early age (usually within the first year of life) recurrent bacterial and opportunistic infections, including Pneumocystis carinii pneumonia and intractable diarrhea due to cryptosporidium infection. Despite substitution treatment with intravenous immunoglobulin, the overall prognosis is rather poor, with a death rate of about 10% before adolescence. Belongs to the tumor necrosis factor family.

The Anti-CD40 [chi220] reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's CD40 can be used in a range of immunoassay formats including, but not limited to, Blocking (BL), Therapeutics. This antibody blocks the interaction between CD40 and gp39. Chi220 was used either alone or in combination with CTLA4-Ig in an immunosuppresive therapy study to check the survival of renal allografts in non human primate models (PMID: 12394833). Chi220 blocks binding of CD154, it also has partial agonist properties and weak stimulatory potential. It synergisticaly works with LEA29Y (belatacept) to prolong the islet allograft survival in a non-human primate Macaca fascicularis (PMID: 15611281). A study on protection of islets xenographs in diabetic macaques used a combination of Chi220 with IL-2 receptor specific antibody Basiliximab for immunosuppression with induction therapy. Chi220 is used as an sCD154 blocking agent and it was seen that it binds sCD154 in a dose dependant manner (PMID: 21920020). Two humanized versions of this antibody were generated F4 and L3.17, also showed similar properties. Humanized versions of this antibody are available on request. Researchers should empirically determine the suitability of the CD40 cd40lg for an application not listed in the data sheet.
